I have a 2005 Highlander and a 2010 Prius. Love them both. My SUV has 150k miles and going strong, Prius has 70k, same. Still going strong (knock on wood!) As much as I love my Highlander, it has not been trouble free. I had to have the brakes completely overhauled to the tune of $3000, and a rear differential went that was another thousand bucks. Sunroof cable has snapped three times (twice under warranty) So far so good with the Prius, ironically bought that one used with 8k miles. Regardless, I would recomend Toyota overall. Previously had a jeep which broke down with 18 miles on it while driving home from the dealer and several Audi's which were great performers but when something goes wrong, it's usually huge and there is nobody in my area that is qualified to work on them, closest dealer is 35 minutes away. Not sure what area of the country you live in or if snow is a factor, prius is a zero in the snow, highlander is better but still not great. Have you looked at KIA or Hyundai? They are supposedly the best and most reliable on the market now.
