Mr. Rebates-Can Anyone Tell Me?

I just signed up with Mr. Rebates recently and I'm not really sure I understand how it works. Do you have to go through the Mr. Rebates website to get to the store and then make your purchase in order for it to show up on your account? Or, can you go directly to the store and make your purchase? If so, how does it link your purchase with Mr. Rebates?
